A new semi-alkaline proteinase produced by Streptomyces cinereoruber.

چکیده
Semi-alkaline proteinases having optimum pH between 8 and 9 of Streptomyces origin. have been reported as products of Streptomyces erythreus,3)Streptomyces fradiae4)and Streptomyces madurae.5)The proteolytic activity of all of the above semi-alkaline proteinases can be inactivated by addition of diisopropylfluorophosphate. We have recently isolated a new semi-alkaline proteinase from the cultured-broth of Streptomyces H 55SY 7 which was isolated from a soil sample collected at Kochi prefecture and identified as Streptomyces cinereoruber.6) The proteolytic activity of the new semi-alkaline proteinase is not inactivated by addition of diisopropylfluorophosphate, but inactivated by addition of ethylenediaminetetraacetate. Production, isolation, purification and enzymatic characteristics of the proteinase are presented in this paper. The semi-alkaline proteinase was produced by cultivation of Streptomyces H 55-SY 7 in shaking flasks containing a medium composed of starch, glucose, soy bean meal, yeast extract and various inorganic salts on a reciprocal shaker. The proteinase was precipitated from the broth filtrate by saturation with ammonium sulfate and the crude enzyme mixture was reprecipitated by addition of cold acetone after dialysis against water. The enzyme was purified by gradient column chromatography on DEAE-cellulose after gel filtration

Characterization of proteolytic enzyme secreted by Streptomyces cinereoruber ssp. cinereoruber isolated from human pleural fluid
Actinomycetes are an uncommon agent of human infections and its pathogenic factors are not known. The present study reports a rare case isolation of an actinomycete from a woman pleural fluid; the strain was identified by 16S rRNA gene sequence analysis. This strain was tested to produce an extracellular protease that hydrolysis gelatin, casein and hemoglobin on agar mediums. The purification o...

متن کاملAntioxidant and cytotoxic activities of metabolites produced by a new marine Streptomyces sp. isolated from the sea cucumber Holothuria leucospilota
Marine microorganisms are important sources for novel natural products. Hence, the aim of this study was to introduce marine microorganisms with the capability of producing antioxidant and cytotoxic metabolites. For this purpose, ten live sea cucumbers (Holothuria leucospilota) were collected from Larak Island, Persian Gulf. Then, their intestine contents were serially diluted and treated by he...

Background: Streptomyces is one of the most important prokaryotic microorganisms, and their secondary metabolites have high antimicrobial and cytotoxic properties. Three-quarters of the antibiotics known to be produced by these bacteria. As a result, finding new effective compounds from these microorganisms can be a way to treat cancer. The aim of this study was to isolate and screen halophilic...
